I'm little confused over construction of a Duplex house vs independent floor house on 30X50 site for a nuclear family (4, husband, wife and 2 kids), appreciate your inputs on this..

Better if you share more details like how many Bedroom, or media specific rooms or additional car parking..etc you are planning for your new house.

A 30X50 plot size can comfortably accommodate a 2.5/3 BHK with a car park.

Going for duplex is better if its only for self use. In case you have plans to rent out any floor in future, you can go for independent floors.

Also remember, maintenance/cleaning is an added issue, if you go for more area, whether its duplex or individual floors.
